The study, published in the Journal of Neuroscience, found that rats given THC had higher levels of activated ERK, displayed more anxiety behaviors, and were more susceptible to fear-based learning. Rats that were given both CBD and THC behaved similarly to control rats: they had normal levels of activated ERK, exhibited less anxiety, and were less sensitive to fear-based learning.

Based on these findings, the researchers propose that CBD inhibits THC’s ability to overstimulate the ERK pathway in the hippocampus, thereby preventing its negative side effects.

“Our findings have important implications for cannabis prescription and long-term cannabis use. For example, individuals who are more susceptible to cannabis-related side effects should limit their use to strains with high CBD and low THC content “Laviolette stated. “More importantly, this discovery paves the way for the development of more effective and safer THC formulations.”

The study’s lead author, Ph.D. candidate, and Vanier Scholar Roger Hudson says another interesting finding was that CBD alone had no effect on the ERK pathway. “CBD had no effect on its own,” he said. “We were able to completely reverse the direction of the change on a molecular level by co-administering CBD and THC. CBD was also capable of reversing the anxiety-like and addictive-like behavior caused by THC.”

Laviolette says they will continue to identify the specific features of this molecular mechanism as part of their research. The study will look into how to make THC with fewer side effects and improve the efficacy of CBD-derived therapies.
